Renee Zellweger immediately comes to mind. She put on something like 30 pounds to play Bridget Jones - which doesn't seem so hard - but then she had to lose it all for Chicago (and other roles).

In fact she did that twice considering the Bridget Jones sequel.

Toni Collette's physique has been through some changes - compare her in Muriel's Wedding to Changing Lanes - but I don't think any of it was done strategically for artistic purposes. I think she is more in the Oprah category where her weight is just a bit out of control.
